Beskrivning 2-Bromohexadecanoic acid (2-BP) är en icke-metaboliserbar analog av palmitat och fungerar som en palmitoyleringshämmare, vilken hämmar multiple palmitoyl-acyl transferase (PAT)-aktivitet mot MyrGCK(NBD) och FarnCNRas(NBD) med identiska IC50-värden på ca 4 µM. 2-Bromohexadecanoic acid kan direkt och irreversibelt hämma palmitoyltransferas-aktiviteten hos alla DHHC (Asp-His-His-Cys)-proteiner.

2-BP (2-Bromohexadecanoic acid) induces the interactions between GSDME-N and GSDME-C, and inhibits the GSDME-C palmitoylation, exhibiting chemotherapy induced pyroptosis.

  • Metod

    Cells are pre-seeded overnight until cell density reached ~60%. The culture medium is switched to fresh medium containing relevant drugs and incubated for a period of time as indicated. After treatment with 2-BP (2-Bromohexadecanoic acid) and TNFα+CHX, Hela cells are collected at the indicated time points and stained with FITC-Annexin V and PI, and subjected to Hoechst 33342 and PI double staining. The culture supernatants are collected to measure the percentage of LDH release.
